He was born in Zalaya, New Spain. As Dean of the Metropolitan Cathedral of Manila, he also held a professorial chair of the Sacred Canons in the University of Santo Tomas. Prior to his becoming Bishop-elect of Cebu, he was Provisor and Vicar General of the Archdiocese cede vacante.
His appointment to Cebu according to letters signed by King Philip V in-cluded also his nomination as Governor and Captain General ad interim of the Philippines and President ad interim of the Royal Audiencia. The letters were dated August 15, 1734 according to the Historia General de Filipinas by Fr. Juan de la Concepcion, volume 12, page 196. They were addressed to Bishop Juan de Arechedera, Bishop-elect of Nueva Segovia.
Bishop Manuel Antonio de Osio was installed in Cebu on April 26, 1733 and died in July 21, 1737.
